Walaupun lahir di Italia, orang tua Cole Sprouse berkebangsaan Amerika.

Ibu Cole Sprouse bernama Melanie Wright dan ayahnya bernama Matthew Sprouse.

Cole Sprouse memiliki saudara kembar bernama Dylan Sprouse yang berusia 15 menit lebih tua darinya.

Saat Cole dan Dylan Sprouse berusia empat bulan, orang tua mereka pindah ke Long Beach, California.

Cole Sprouse memulai kariernya di dunia seni peran bersama saudara kembarnya, Dylan Sprouse saat masih berusia 6 bulan.

Pada 1993-1998, Cole Sprouse bersama saudara kembarnya berbagi peran sebagai Patrick Kelly di serial ABC berjudul 'Grace Under Fire (1993)' dan Julian dalam 'Big Daddy (2000)'.

Cole Sprouse pertama kali muncul tanpa saudaranya dalam film serial NBC berjudul 'Friends (1994)' sebagai putra Ross Ben Gellar.

Pada musim gugur 2003, mereka berdua kembali berbagi peran sebagai pemeran utama Jeremiah dalam 'The Heart Is Deceitful Above All Things (2004)'.

Pada 2005, Cole Sprouse bersama saudaranya membintangi sebuah sitcom (komedi) milik Disney Channel berjudul 'The Suite Life of Zack & Cody (2005)'.

Setelah enam tahun membintangi program ini, Cole Sprouse memilih melanjutkan pendidikannya di New York University dan vakum dari dunia hiburan.

Cole Sprouse pernah dinominasikan sebagai MVP (Most Valuable Player) dalam minggu keenam Disney Channel Games 2007 memimpin tim biru untuk Super Soccer namun tidak berhasil menang.

Cole Sprouse kalah dari rekannya di 'The Suite Life of Zack and Cody', Ashley Tisdale yang berada di tim merah.

Saat masih menjadi mahasiswa di NYU, Cole Sprouse pernah bekerja sebagai event photographer untuk Meltdown Comics di Los Angeles, California pada 2011.

Saudara kembar Dylan Sprouse ini kembali ke dunia hiburan setelah menyelesaikan studinya pada 2015 dengan membintangi Jughead Jones dalam serial CW 'Riverdale (2017)'.

Peran Cole Sprouse sebagai Jughead menjadi favorit para penggemar 'Riverdale'.

Kemudian Cole Sprouse semakin dikenal setelah membintangi Will Newman, seorang remaja yang mengidap penyakit langka cystic fibrosis dalam film box office 'Five Feet Apart (2019)'.

'Five Feet Apart (2019)' menjadi salah satu film bergenre remaja yang paling laris dari tahun 2010an.

Selain aktif sebagai aktor, Cole Sprouse menyukai fotografi.

Cole Sprouse pernah memotret untuk sampul majalah dan muncul di The Sunday Times dan L'Uomo Vogue.
